NFκB-IκB-GFP
NF-κB Binding Element can recognize and bind with the free NF-κB. NF-κB regulates the expression of downstream genes by binding with the element. IκB is the inhibitor of NF-κB. The activity of NF-κB will be large-sized decreased, when IκB binds with NF-κB. The green fluorescent protein (GFP) is a protein composed of 238 amino acid residues (26.9 kDa) that exhibits bright green fluorescence when exposed to light in the blue to ultraviolet range. We fuse the fragments of IκB and GFP. The two proteins can co-express. We combined these three sequences(NF-κB, IκB, GFP) into a specific sequence by using overlap PCR method, thus, the recombinant fragment could provides its significant bio-effects.
IκB fragment has two PstI restriction sites. We tried to use point mutation method to mutant a A into T of PstI, but it hardly work because of its functionality lost.
